Rap1GAP is expressed in the absence of methylation at histone (Histone 3 at lysine 27 or H3K27) and promoter hypermethylation (left panel). EZH2, the catalytic component of the PRC2 complex, di- and tri-methylates H3K27 resulting in chromatin compaction, the mark of repression. Consequently, Rap1GAP is silenced (middle panel). CpG islands in the promoter region of Rap1GAP are further methylated by DNA methyl transferase (DNMT) recruited by PRC2 complex. The coordination of these enzymes results in further repression and epigenetic silencing of Rap1GAP (right panel). Methylation and consequent silencing are denoted by red.
