Densitometry assays of GTP-cdc42 after PI3K modulating treatment in Caco-2 clones. EGF enhanced while wortmannin (wtm) treatment suppressed cdc42 activation in both cell types. Treatment effects were greater in Caco-2 cells (Control vs EGF vs wtm vs EGF+wtm = 0.69 ± 0.08 vs 1.6 ± 0.23 vs 0.39 ± 0.04 vs 0.47 ± 0.05 in Caco-2 cells and 0.21 ± 0.05 vs 0.72 ± 0.14 vs 0.13 ± 0.02 vs 0.2 ± 0.05 in Caco-2 ShPTEN cells; p<0.001 for effects of treatment and cell type on cdc42 activation;Two way ANOVA; n=3).
