Eukaryotic argonaute proteins are characterized by having both PAZ and Piwi domains. Piwi domains are found only in Ago proteins and are structurally related to the RNase H family of ribonucleases. Nucleic acid binding sites are indicated by vertical lines. The 5′ ends of siRNAs and miRNAs are important for mRNA target recognition and definition of the site of RNA cleavage. These binding sites are indicated by grey ovals. Active sites (black ovals) in the Ago1 ribonuclease correspond to Tyr684, Glu686, Pro757 and Gly895 and in Ago 2 correspond to Asp740, His742, Asp812, and His950.
