Analysis of the effect of ac34 deletion on viral gene expression. (A) Time course analysis of IE1, GP64, 38K, P6.9, and VP39 synthesis. Sf9 cells were infected with vAcWT or vAc34KO at an MOI of 1 TCID50/cell and harvested at different time points. The cell lysates were subjected to immunoblotting with anti-IE1, anti-GP64, anti-38K, anti-P6.9, or anti-VP39 antibody. An anti-actin antibody was used as a loading control. (B and C) qRT-PCR analysis of viral gene transcription. The effects of ac34 deletion on the transcription of several genes were measured with qRT-PCR at 12 (B) and 24 (C) h p.i. The transcript level of each gene was normalized to the genome copy number. The effect of ac34 deletion on gene transcription is shown for each gene as the percentage of detected transcripts relative to vAcWT. The error bars represent the standard deviations from the mean.
