VcHfq preferentially binds Qrr1 with its proximal face. Representative EMSAs for a distal face mutant (Y25D), a proximal face mutant (K56A) and a distal and proximal face double mutant (Y25D K56A) of VcHfq binding to Qrr1 sRNA. Qrr1 was labelled with 32P and was present at 5 nM in each lane and the concentration of hexameric VcHfq protein is specified above the lane. The mobilities of free Qrr1 RNA, the 1:1 VcHfq:Qrr1 complex and higher order complexes are indicated on the right of the gels. The dissociation constant (Kd) for the 1:1 VcHfq:Qrr1 complex, calculated from three repeats, is reported above each gel.
