Time-dependent increase of H2DCF oxidation in dye-loaded U937 and Jurkat cells, and the lack of dye photoactivation by repeated measurements with the plate reader. Identical cell samples were loaded in different wells of a 96-well plate; DCF emission was measured once or several times during 1-hr incubation (mean +/− s.e.). The stopwatch was started (0 min) when all cell samples were dispensed in the respective wells of a 96-well plate. Note gradual increase in DCF formation due to cell metabolism or self-oxidation of H2DCF, whereas repeated measurements of the same well did not cause additional oxidation. See text for more detail.
