PDZ-RGS3 interacted with GSK3β but not Dvl. A, PDZ-RGS3 was co-immunoprecipitated with GSK3β. HEK 293 cells transiently transfected with Au1-tagged PDZ-RGS3 were treated with Wnt3a (20 ng/ml) or not for 15 min. The lysates were used for immunoprecipitation and immunoblots with the indicated antibodies. B, Wnt3a increased the endogenous GSK3β and PDZ-RGS3 interaction, but reduced the GSK3β and Dvl1 interaction. HEK 293 cells were treated with Wnt3a ligand (20 ng/ml) for 15 min. The cells were lysed and GSK3β immunoprecipitated. The indicated proteins were detected by immunoblotting.
