Cytokine profiling from DCs infected with H37Rv or mutant strains.
A, measurements of levels of IL-12p40 and IL-1β from supernatants of DCs infected with H37Rv or mutant strains (H37RvΔRD1, H37RvΔESAT-6, or ESAT-6-complemented H37RvΔESAT-6) of H37Rv at different points. B, levels of IFN-γ and IL-4 from the culture supernatants of DCs infected with H37Rv or mutants (H37RvΔRD1, H37RvΔESAT-6, or ESAT-6-complemented H37RvΔESAT-6). H37Rv and ESAT-6-complemented H37RvΔESAT-6 (Comp ESAT-6) direct differentiation of both Th1 (IL-12p40 and IFN-γ) and Th2 (IL-4) cytokine responses as well as IL-1β production, whereas H37RvΔRD1 and H37RvΔESAT-6 induce only Th1 cytokines in infected DCs co-cultured with naïve OT-II CD4+ T cells. IL-4 and IL-1β levels were significantly increased (p < 0.01) in H37Rv and Comp-ESAT-6 infected DCs compared with DCs infected with the other strains.
