Overexpression of AhMGAT increases TAG formation. MGAT was overexpressed in wild-type yeast in the presence of [14C]acetate (0.5 μCi mL−1). The yeast cells were grown in SM-U containing 2% Gal for 24 h, and equal absorbance (A600 = 20) of cells was harvested and lipids extracted using chloroform:methanol (1:2, v/v). The neutral lipids and phospholipids were separated by silica-TLC, and the radioactivity was quantified by a liquid scintillation counter.
