Non-canonical Wnt7a does not bind heparin. A, 293T cells transfected with empty vector (Ctrl), Sulf2-Myc, or inactive Sulf2(C-S)-myc expression vector were assayed for Sulf2 expression by Western blot using the Myc antibody. Wnt3a (1 ng) bound to heparin or Sulf2-digested heparin was assayed. Sulf2 reduced Wnt3a-heparin binding. B, increasing amounts of Wnt7a (0, 10, 30, or 100 ng) were incubated with heparin or Sulf2-digested heparin to allow binding. The amount of Wnt7a bound to heparin was assayed by Western blot.
