The PDZ domain in CNK3 is essential for its ability to interact with ENaC and the key ENaC-regulatory kinase, SGK1. HEK 293T cells were transiently transfected with FLAG-tagged α-, β-, and γ-mENaC subunits and V5-tagged WT- or ΔPDZ-CNK3 with or without SGK1 as specified. Shown are co-immunoprecipitation assays demonstrating the drastically reduced ability of ΔPDZ-CNK3 to interact with ENaC (A) (n = 4) or SGK1 (B) (n = 5).
