Loss of membrane asymmetry induced by IAPP measured by the pyrene-PC assay. Lipid translocation induced by 1 µM monomeric IAPP, 1 µM monomeric IAPP in the presence of 1 µM preformed IAPP fibers, and 0.5 µM MSI 78 in 20 µM (A) POPC/POPS 7/3 or (B) POPE/POPC/POPS 3/4/3 large unilamellar vesicles labeled with 3% PyPC in the outer leaflet. Both peptides were added at the 10 minute mark. All experiments were performed at 37°C in 10 mM phosphate buffer, 100 mM NaCl, pH7.4.
