Memory Phenotype. Homologous prime boost regimens favor a Tcm cell phenotype in the peripheral blood of Ad5 immmune mice. Memory phenotype was defined as percent of CSP (NYD) tetramer positive cells that are Tem cells (CD62Llo CD127+) and percentage that are Tcm cells (CD62L+ CD127+) as opposed to percent of Tem and Tcm cells that are Tet+. PBMCs were collected on day 14 post final injection and stained according to the above defined memory phenpotype. Example of gating appears above the graphs (A). Percentage of Tcm Cells was significantly higher PBMCs from homologously boosted animals in Ad5 immune mice (B). There was no significant difference in the percentage of Tem cells present between any of the groups (C). Bars represent ± standard error. Statistical analysis was completed using One Way ANOVA with Student-Newman-Keuls post-hoc Test.
