ATP7BR875,S340/341A and ATP7BR875,S340/341G behave as ATP7BR875 in a copper-saturated state. Hek293TRex cells transiently transfected with Flag-ATP7BR875, Flag-ATP7BR875,S340/341A, or Flag-ATP7BR875,S340/341G were kept in basal media, copper depleted using 100 μm BCS for 2 h or treated with 100 μm CuCl2 for 2 h. Cells were immunostained using Flag antibody (green) and TGN 38 (red); merged images are shown (Scale bar: 5 μm). A, Flag-ATP7BR875 is localized in ER in basal (left panel) and copper-depleted conditions (middle panel), and moves to vesicles upon copper treatment (right panel).
