Single-turnover unwinding kinetics at different ATP concentrations. A total of 2 nM DNA substrate (16-bp duplex with a 15-nt 3′ tail) with 30 nM BLM, ATP of varying concentrations with 2.5 µM dA8 and 2 µM DNA trap, and 5 µM dT56 were first preincubated, respectively, and the reaction was initiated by rapid mixing of the three solutions.
