Gate strategies used to determine the expression of Toll-like receptor (TLR)2 and TLR4 on monocytes, myeloid dendritic receptors (mDCs), and plasmacytoid dendritic receptors (pDCs). Mononuclear cells were gated according to physical characteristics, excluding dead cells. Monocytes were then gated as CD14⁺ cells; mDC as Lin 1⁻ CD11c^high; and pDC in two ways, BDCA-2⁺ CD123^high or Lin 1⁻ CD123^high. Each specific subpopulation was plotted as histograms to show the expression of TLR2 and TLR4. Data are presented as overall mean fluorescence intensity (MFI), after subtraction of isotype staining background.
