CD80 expression is increased in pDCs exposed in vitro to HIV-1. CD80 expression was measured by flow cytometry in total PBMCs exposed in vitro to HIV-1_{H9 HTLV-IIIcc} for 2 h before stimulation with 10 nM Pam₃CSK₄ and 10 ng/ml LPS for 18 h at 37°C and 5% CO₂. Mononuclear cells were gated according to physical characteristics excluding dead cells, and pDCs were then gated as Lin 1^– CD123^high cells. Representative results of three independent experiments are shown as median and range of the fold change in the expression of the marker [mean fluorescent intensity (MFI) in overall subpopulation, after isotype control subtraction] compared to the control without infection or TLR stimulation.
