(A) Northern blot analysis of miR169 in response to Pi availability (+P, 1 mM and −P, 5 µM) in 8 and 14-day-old hen 1-1 and in WT seedlings. miR156 probe was used as a control to confirm that mature microRNAs in general have decreased levels or are absent in hen1-1. U6 probe was used as a loading control. Ethidium bromide-stained tRNA and rRNA is shown below the blots to indicate the relative amount of total low molecular weight RNA loaded per lane. (B) and (C) Tissue specific expression directed by the NF-YA10 (B) and miR169ij (C) promoters as revealed by GUS histochemical assays in seedlings grown in Pi contrasting conditions 14 days after germination (dag). CL, SAM, TL, HYP, R, LR and RT, are cotyledonary leaf, shoot apical meristem, true leaf, hypocotyl, root, lateral root and root tip, respectively.
