The CCL2 response to Pneumocystis in primary alveolar epithelial cells is dependent on IL-1R. Primary murine AECs were isolated from C57BL/6 WT and IL-1R−/− mice. Confluent monolayers were exposed to freshly isolated murine Pneumocystis at cyst-to-AEC ratios of 0.5:1, 1:1, 3:1, and 4:1. AECs were also treated with either medium alone (control), TNF-α (10 ng/ml), or IL-1β (10 ng/ml). CCL2 levels in the supernatants at 6 h were measured by ELISA.
