U20 reverses HHV-6B-induced resistance to apoptosis. (A) HCT116 wt and U20-S cells were transfected with U20 siRNA (200 nM) or scrambled (Scr.) siRNA (200 nM) for 48 h and incubated in the presence or absence of TNF-α (25 ng/ml) and CHX (10 μM) for 4 h, followed by Western blot analyses with antibodies against PARP or GAPDH (loading control). (B) Quantification of cleaved PARP from panel A. (C) HCT116 cells were transfected with U20 siRNA (200 nM) or scrambled siRNA (200 nM) for 24 h and mock treated or HHV-6B infected for 48 h and then incubated in the presence or absence of TNF-α and CHX for 4 h, followed by Western blot analyses with antibodies against PARP, GAPDH (loading control), or 7C7 (infection control). (D) Quantification of cleaved PARP from panel C. Representatives of at least two experiments are shown.
