Schematic diagram of sequential rearrangement steps at the TCRβ locus. Two successive genetic rearrangements occur at the β- chain. The rearrangement of the TCRβD to TCRβJ segments occurs first and generates a by-product, a specific DJβTREC. This is followed by the recombination of the V to DJ segments, which also generates a specific VDβTREC. These two types of TREC molecules can be quantified, but there are too many possibilities with 65 Vβ segments to consider all rearrangements.
