TGIF1 silencing limits the radiation- and TNF-α-induced proinflammatory state of endothelial cells. A, HUVECs were transfected for 24 h with nontargeting siRNA or siRNA targeting TGIF1. Silencing efficiency was measured at mRNA and protein levels 24 h after transfection. B, HUVECs were transfected with nontargeting siRNA or siRNA targeting TGIF1 and exposed or not to 10-Gy irradiation. mRNA expression levels of IL-6, IL-8, CXCL1, MIP-2, and MCP-1 were measured 3 h after irradiation. C, HUVECs were transfected with nontargeting siRNA or siRNA targeting TGIF1 and treated or not with 0.1 ng/ml TNF-α. mRNA expression levels were measured 3 h after incubation.
