Sequence alignment of various GlyR intracellular TM3-4 loop sequences between amino acids 337 and 433. The amino acid numbering refers to the immature polypeptide, modified from Melzer et al. (15). The length of this loop differs between GlyRα variants due to alternative splicing within the sequence. The sequences of the human (H. sapiens; hs) and mouse (M. musculus; mm) TM3-4 loops of the GlyR α1, α2, and α3 subunits (including different splice variants) are shown. The alignment is arbitrarily subdivided into conserved (C1–C3) and variable regions (V1 and V2, shaded in gray). Most charged amino acids (+, positively; −, negatively) are conserved between all variants.
