MNA amplified and MYCN overexpressing cells are characterized by a delicate equilibrium between pro- and anti-apoptotic factors of the p53 pathway that are directly or indirectly regulated by MYCN. Unbalancing these factors may easily trigger apoptosis in this context. In particular Nut-3 is highly effective because it increases p53 expression and its S⁴⁶ phosphorylation via HIPK2 induction and at the same time it represses Gal-3, in MNA NB cells.
