DBC1 did not affect SIRT1 activity in vitro in liver cancer cells. A549 and SNU-182 (A and B) cells were transfected with siRNAs targeting SIRT1 or DBC1 (50 or 200 nM). Etoposide (20 μM) was administered for 12 h to induce p53 hyperacetylation due to DNA damage. Knockdown of SIRT1 or DBC1 and acetylation of p53 were evaluated by western blotting with the indicated antibodies. All membranes were probed for GAPDH to confirm equal protein loading.
