Mast cell degranulation response to stimulation by HTAs. The mast cells were primed with 100% IgEDNP in (A), and 25% IgEDNP and 75% orthogonal IgE (IgEdansyl) in (B), then exposed to one of the eight HTAs at the indicated concentrations. The potency, both in terms of concentration required to stimulate the maximum response as well as the overall strength of degranulation response, directly correlated with the monovalent hapten affinity. (C) The effect of hapten specific IgE (IgEDNP) percent for achieving maximum degranulation response was investigated. The maximum response for all HTAs was observed for RBL cells primed with 25% IgEDNP.
