(A) In resting cells BIN2 is bound to RIN3 at a proposed BIN2 Binding Domain (BBD). (B) SCF binding to KIT stimulates RAS. RAS activates RIN3, though a RAS Association (RA) domain, and other effectors (RAF, PI3K). RIN3 releases BIN2, which might then become available for membrane association. The RIN3 GEF domain activates RAB5 to promote early endosome fusion and trafficking. Internalized KIT may continue signaling, be recycled to the plasma membrane, or be targeted for degradation.
