Combination of tamoxifen and Herceptin improves the sensitivity of BT/HerR cells to Herceptin. (A) BT and BT/HerR1.0 cells were deprived of steroid for 72h and then incubated in the presence of 1 nM 17β-estradiol (E2) with increasing concentrations of Herceptin (HPT) or its combination with 1 μM tamoxifen (Tam) for 6 days. Cell proliferation was expressed relative to optical density at 490 nm of each cell line in the presence of 17β-estradiol. *, P<0.05, combined agents versus Herceptin alone or tamoxifen alone.(B) BT and BT/HerR1.0 cells were deprived of steroid for 72h and serum starved for 24h, and then treated with 1 μM Herceptin alone or its combination with 1μM tamoxifen for 24h. Levels of total (t) and phosphorylated (p) MAPK, AKT and ERα at Ser118 were measured in cellular extracts by Western analysis. β-actin was used as a control for equal loading and transfer.
