FISH confirmation of the breakpoint. A. Cartoon of breakpoint and BAC probes used for FISH. The breakpoints are designated by arrowheads and genome coordinates for each chromosome. The Ts65Dn marker chromosome contains proximal Chr 17 and distal Chr 16 sequences as indicated in red. Bacterial artificial chromosomes from the RP23 library that contain genomic sequences flanking the breakpoints are depicted as colored boxes with the colors corresponding to the fluorophores used to label these clones. The letters within the boxes indicate which pairs of probes were used to generate the images below. The IDs of the clones are also noted. B. Hybridization with probes RP23-325G3 and -67A9 showed that marker chromosome contains sequences from the proximal Chr 17 but not Chr 16 probe. C. Hybridization with probes RP23-18M23 and -178A3 revealed that the marker chromosome includes sequences from the distal Chr 16 but not Chr 17 probe.
