Schematic illustrating the synthetic approach used to prepare gadolinium (Gd)-labeled, polydisulfide dendrimer nanoclusters (DNCs). PAMAM dendrimers(G3) were first reacted with SAT(PEG)4. The thiolated dendrimers were then suspended in an alkaline buffer for deprotection and to initiate the formation of polydisulfide DNCs. Once DNCs with a desirable hydrodynamic diameter were formed, free thiols were quenched with maleimide and the remaining amines were functionalized with [Gd-C-DOTA]-1.
