Endogenous Orai1 protein is expressed as two different isoforms in HEK293 cells. Western blot showing the expression of Orai1 protein in HEK293 cells transfected with negative control siRNA (siControl) or siRNA against human Orai1. Cells were lysed in RIPA buffer. After overnight treatment with PNGaseF, samples were dissolved by 4–20% gradient SDS-PAGE and analyzed by Western blotting using an anti-Orai1 antibody. Actin expression confirms that similar amounts of protein were loaded in each lane. Representative blot from three independent experiments is shown.
