Mitochondrial ATP production.
A, intact mitochondria were isolated from wild type PCF T. brucei (WT) and from non-induced (−tet) and tetracycline-induced (+tet) ΔTbMCP5/TbMCP5-nmycti (conditional double knock-out (cdKO)) trypanosomes at 72 h. Mitochondrial ATP production was initiated by the addition of succinate (left) or α-ketoglutarate (right). ATP production in WT mitochondria from succinate or α-ketoglutarate was set to 100%. Mitochondrial ATP production (export) was completely inhibited by the addition of CATR. Data shown are means of more than three independent experiments. B, proline consumption and product formation (succinate, acetate) for wild type and non-induced ΔTbMCP5/TbMCP5-nmycti (TbMCP5-depleted) PCF T. brucei, cultured in glucose-depleted SDM80 medium for 72 h. Black and gray bars represent substrate consumption and product formation values for wild type and TbMCP5-depleted trypanosomes, respectively. Data means are derived from more than four independent experiments. Error bars, S.D.
