Calpain inhibition induces coilin foci and alters cleaved coilin fragment levels. (A and B) ALLN treatment induces coilin foci in U2OS and WI-38 cells. U2OS and WI-38 cells were treated with ALLN or DMSO (vehicle) for 16 hr. The treated cells were fixed and immunostained for coilin (green), SMN (red) and nuclei (DAPI) (blue). Coilin, SMN and DAPI images were overlaid in merge panel. In U2OS and WI-38 cells, arrows indicate cells with predominantly nucleoplasmic coilin, arrowheads indicate coilin foci with SMN (Cajal body) and double arrowheads mark coilin foci without SMN (A).
