Validation of MRM. (A) Representative extracted ion chromatograms for three transitions 285.1/183.1, 285.1/211.2 and 285.1/188.1 obtained from a sample containing 1 pg of chemically synthesized MIF-1. MIF-1 (100 fg–10 ng) was loaded onto the chromatography column followed by MRM analysis to determine the limit of quantification, limit of detection, and linearity response of the assay. (B) The assay was linear over four orders of magnitude and the limits of quantification and detection were determined to be 300 fg and 60 fg, respectively. The error bars in (B) represent the standard deviation of the mean and in some cases are equal to or smaller than the size of the symbol.
