A. 293-Kb cells were radiolabeled with [35S]-Met for 10 minutes in the presence of varying concentrations (µM) of CHX and puro, followed by precipitation of polypeptides with trichloroacetic acid (TCA). TCA precipitates were solubilized and radiolabel incorporation was measured by liquid scintillation counting. [35S] signal was normalized to untreated controls (n = 3; mean ± s.e.m.) B. Separation of TCA precipitates from samples in A by tricine SDS-PAGE in 10% gels. Gels were dried and exposed to a PhosphorImager plate overnight. Results are representative of three independent experiments.
