NRK cells of indicated cell cycle phases were double stained by the following antibodies: (A) LX108 (T220/T224) and R3G3 (S277); (B) LX108 and RB7 (S376); (C) a polyclonal antibody pS277 that recognized phosphorylated S277 and RB7. Cell cycle stage was determined by the morphology of the DNA. Note that the signals for R3G3 and RB7 were detected earlier than that of LX108, indicating that the three recognized sites are activated sequentially during mitosis. The intensity of the LX108 signal correlated with the extent of Golgi fragmentation, suggesting that T220/T224 phosphorylation is critical for mitotic Golgi disassembly.
