Influence of gapped DNA on bypass of a minor groove DNA lesion. (A) Replication on an unadducted template. The efficiency of bypass and the measurement of incorporation beyond the control dG templating base were calculated at different dCTP concentrations in the absence or presence of a downstream strand (square and triangle symbols, respectively). Bypass efficiency was calculated as outlined in Experimental Procedures. All reactions included 10 fmol of pol β and were carried out for 5 min. (B) Bypass with a C10S BPDE-dG adducted template. (C) Bypass with a C10R BPDE-dG adducted template. Note that bypass efficiencies in the absence of a downstream oligonucleotide are so poor that those values do not reflect a full bypass.
