DNA sliding increases 5-meC excision efficiency. Purified proteins (20 nM) were incubated at 30°C with fluorescein-labelled substrates S and SL1-2 (20 nM) containing a single 5-meC:G pair. Reaction products were separated in a 12% denaturing polyacrylamide gel and quantified by fluorescence scanning. The relative processing efficiencies (E_rel) were determined in kinetic assays as described in the experimental procedures (see Supplementary Table S2). Values are mean ± SE from two independent experiments, adjusted with the unbiased estimator described in (36).
