The fold improvement was calculated as (strain performance)/(EcNR2 performance), where performance refers to the average number of allele conversions per clone, or the fraction of clones with 5+ or 0 conversions. These metrics were the average of individual metrics for Oligo Sets 1, 2 and 3. In all three categories, Nuc5-.dnaG.Q576A exhibited an effect that was roughly an additive combination of the effects yielded in Nuc5- and EcNR2.dnaG.Q576A.
