The diagram of Pih1 regions and motifs that are involved in R2TP complex formation. Pih1 is represented by a rectangle from amino acid 1 to amino acid 344. The Pih1 fragments used in this study were color coded as Pih11–230 (blue), Pih1231–284 (orange), and Pih1285–344 (yellow). It should be noted that fusion protein GFP-Pih1282–344 used in this study contains Pih1 fragment Pih1282–344 that starts two amino acids before Leu284. The two predicted intrinsically disordered regions IDR1 (Gly195–Leu227) and IDR2 (Asn240–Tyr267) are also highlighted. The five hydrophobic amino acids clusters in the Pih1 C terminus are indicated as gray bars, and the E236 is indicated as black bar. The overall sequence fragments required for Rvb1/Rvb2 and Tah1 bindings are indicated on the top of the Pih1.
