Axl up-regulation by mutant p53 has physiological significance. (A, C) Growth rate of H1048 and H1793 lung cancer cells depends on the Axl level. H1048 and H1793 cells were transfected with control or Axl-specific siRNA, plated in equal numbers, and harvested each day for 5 days to determine the rate of doubling. In parallel, p53, Axl, and Erk2 levels were determined by immunoblotting (right). (B, D) Motility of H1048 and H1793 cells depends on the Axl level. H1048 and H1793 cells were transfected as above. Forty-eight hours later, standard scratch assays were carried out as described in Materials and Methods and migrating cells stained and counted. Bar = ±1 standard deviation.
