Nucleotide agonists at the human P2Y12. (A) the human P2Y12 was transformed into yeast cells and incubated with different concentrations of P2Y12 agonists. Receptor activation-dependent growth was measured as OD600 nm after 24 hours. Data are given as mean ± S.D. of three independent experiments, all performed in triplicate. (B) for label-free measurements of receptor activation, a dynamic mass redistribution assay (Epic Biosensor Measurements) with stably transfected HEK cells was performed essentially as described previously (Schroder et al., 2010). The response is shown 6 minutes after compound application. The response of each compound at nontransfected HEK cells was subtracted from the respective response at P2Y12 transfected HEK cells. Data are presented as mean ± S.E.M. of two independent experiments, each carried out in triplicate.
