Fig. 6. Strain limits a committed adipogenic population induced by rosiglitazone.

Fig. 6

(A) mdMSC containing the aP2-GFP construct were cultured in adipogenic medium containing 5 μM rosiglitazone and exposed to a daily strain regimen × 4 d. Total cellular proteins were immunoblotted for designated proteins. (B) Densitometric analyses of PPARγ2, aP2, and adiponectin were performed (n = 3 experiments) after treatment as in (A). * p<0.05, ** p<0.01 shows significant difference from unstrained control. (C) mdMSC treated as in (A) were evaluated by flow analysis. Histograms (left) for aP2-GFP expression in unstrained, control cultures or cultures subjected to a daily strain regimen. Left vertical line indicates threshold for FITC intensity corresponding to positive GFP expression and right vertical line (U20) indicates threshold for FITC intensity corresponding to upper quintile of GFP expression in control cultures. Histogram data were analyzed (n = 3 experiments) for median GFP intensity (top right) and % GFP positive cell fraction corresponding to each threshold level (bottom right). ** p<0.01 shows significant difference from unstrained control.
