MdfA overexpression corrects the cell division defect and restores growth to BC202 at 43°C. (A and B) Normal cell division is restored to BC202 at 30°C in the presence of 0.2% arabinose (B) which induces MdfA expression from the plasmid. (C and D) BC202 harboring empty vector pBADhisA or vector pBADMdfA was streaked on LB-Amp-agar plates in the absence (C) or presence (D) of 0.1% arabinose and incubated overnight at 43°C. BC202 harboring pBADhisA (right sides of plates) fails to grow on either plate. BC202 harboring pBADMdfA (left sides of plates) grows only in the presence of 0.1% arabinose. (E) Western blot using anti-His tag antibody showing MdfA overexpression in BC202 from pBADMdfA in an arabinose-dependent manner. Equal amounts of membrane protein were loaded in each lane.
