Screening analyses indicate that Zscan4 markedly decreases the DNA damage response and increases reprogramming efficiency. (A) The total phospho-H2AX (γ-H2AX) protein levels were markedly decreased when Zscan4f, Dppa3 and NPM2 were used in combination with the Yamanaka factors. Total protein was extracted from reprogramming cells collected on day 4 after viral transfection. β-actin was used as a loading control. (B) The reprogramming efficiency was significantly increased when Zscan4f was used in combination with OSKM or OSK.
