Chondroadherin binding to membrane proteoglycans on 105kc cells. Biotin-labeled membrane proteins (from 20 × 10⁶ cells) were passed over a control agarose column followed by a chondroadherin-agarose column (Mini-Leak). The control (lanes 1, 2, 5, and 6) and chondroadherin (lanes 3, 4, 7, and 8) columns were washed and eluted with 10 mm EDTA containing 6 m urea (lanes 1, 3, 5, and 7) and then 0.5 m NaCl (lanes 2, 4, 6, and 8). Eluted material was precipitated with ethanol and separated on a 4–12% SDS-polyacrylamide gel, electrotransferred to a PVDF membrane, and detected by Western blotting before (lanes 5–8) or after heparitinase and chondroitinase ABC treatment (lanes 1–4). Biotin-labeled proteins were detected using streptavidin HRP and the ECL system.
