Adoptive transfer of wild-type IM into DT-treated chimeric mice restores the PPS14- and PC-specific IgM and IgG responses. (A) Spleen cells from WT FVB mice were enriched for CD11b+ cells using magnetic cell sorting (left panel) followed by purification of CD11b+Ly6Chi cells by electronic cell sorting (right panel) [>99% purity]. (B) Purified CD11b+Ly6Chi cells (2 × 105 cells/mouse) were injected i.v. into CD11b-DTR BM-chimeric mice 24 h after DT injection. DT was administered every 2 to 3 d for the duration of the experiment. DT-treated chimeric mice without injected CD11b+Ly6Chi cells and chimeric mice without injection of DT or CD11b+Ly6Chi cells were included as controls. All groups consisted of seven mice each. Serum titers of Ag-specific IgM and IgG were measured by ELISA. One of two representative experiments. *p ≤ 0.05.
