The V656L mutant increases resistance to cyh. We constructed V656L in pSS607 and placed it in R-1 (Δpdr5 strain) by yeast transformation. Following this, the original chromosomal mutation and the plasmid reconstruction were compared to WT and Δpdr5 isogenic controls for their relative resistances to cyh in liquid culture as described in Materials and Methods.
