Figure 4. — Ribosome subunit analysis of tc-GAR1 rrp8-ΔC and rrp8-ΔC tc-GAR1. Sucrose gradient centrifugation was performed to analyse the influence of conditional expression of Gar1 in combination with rrp8-ΔC on ribosomal subunits. Cells were grown in liquid media without (A) or with 250 μM (B) tetracycline to an OD₆₀₀ of 1. The volume of cell extracts containing 5-OD₂₅₄ units was layered on 10–50% sucrose gradients and was centrifuged for 17 h at 24 500 r.p.m. and 4°C with a SW40 Ti rotor.
