Effect of Cys mutations on Rep68 wt aggregation. Three Rep68 Cys mutants were evaluated by gel filtration chromatography: Rep68 C151S, Rep68 C405S, and a double Cys mutant, Rep68 C151A and C405S. A Superose 6 10/300 GL column was used with a flow rate of 0.5 ml/min. Protein elution was followed by UV detection at 280 nm. When the protein contains the Cys151S mutation (A), no aggregation was observed. In contrast, the C405S mutation still shows an aggregation peak eluting at the void volume of the column (B). (C) The double mutant behaves like the single C151S mutant. The concentration of the proteins was 32.65 μM (2 mg/ml).
